Spend the day in Aquinnah, shops and food at the cliffs and walk to the lighthouse for gorgeous views. Also a beautiful public beach under the cliffs if you pay to park in the lot.

Take a drive "up island" - I would recommend getting in the car to explore Chilmark, the Gayhead lighthouse and Menemsha. The drive is so gorgeous!!
